I have just ahd my car resprayed apart from the area directly surrounding the headlamp.
How can I remove the perspex and, more importantly, replace it with a weathertight seal?

You'll have to remove the whole headlamp - the plexi-glass is bonded to the headlamp case.
IIRC you have to remove the front wing to get to one of the bolts!
Undo the four bolts along the top of the wing and you should be able to slide the wing off forwards (if you have side repeaters - remember to undo them!)
With the wing off, you should be able to access all the bolts to remove the lamp (or if it is the wing you're having sprayed - get this done while it is off to avoid removing the headlamp!)

Here's a tip to go with Bruce's suggestion. Before removing the front wing by sliding forward it would be a good idea to put masking tape on the top edge of the headlamp glass then there is a reduced chance of chipping the paint on the wing edge ajacent to the headlamp. Of course it is easier to get at the headlamp fixings with the front undertray removed. Good luck!
